چکیده

Acute liver injury in its terminal phase trigger systemic inflammatory response syndrome with multiple organ failure. An uncontrolled reaction is difficult to treat and contributes high mortality. Therefore, solve this problem a search for new therapeutic approaches remains urgent. This study aimed explore the protective effects of M. edulis hydrolysate (N2-01) against Lipopolysaccharide-D-Galactosamine (LPS/D-GalN)-induced murine acute injure underlying mechanisms. N2-01 analysis, using Liquid Chromatography Mass Spectrometry (LCMS) metabolomic proteomic platforms, confirmed composition, molecular-weight distribution, reproducibility between manufactured batches. efficiently protected mice LPS/D-GalN-induced injury. The most prominent result (100% survival rate) was obtained by constant subcutaneous administration small doses drug. decreased Vascular Cell Adhesion Molecule-1 (VCAM-1) expression from 4.648 ± 0.445 1.503 0.091 Mean Fluorescence Intensity (MFI) Interleukin-6 (IL-6) production activated Human Umbilical Vein Endothelial Cells (HUVECs) 7.473 0.666 2.980 0.130 ng/ml vitro . drug increased Nitric Oxide (NO) HUVECs 27.203 2.890 69.200 4.716 MFI but significantly inducible Synthase (iNOS) 24.030 2.776 15.300 1.290 NO peritoneal lavage cells 6.777 0.373 µm 2.175 0.279 µm. capability preparation enhance endothelium barrier function reduce vascular permeability Electrical Cell-substrate Impedance Sensor (ECIS) test Miles assay vivo These results suggest as promising agent treating wide range conditions associated inflammation endothelial dysfunction.

Protective effects of Asiaticoside on acute liver injury induced by lipopolysaccharide/D-galactosamine in mice.

Asiaticoside (AS), a triterpenoid product isolated from Centella asiatica, has been described to exhibit anti-in fl ammatory activities in several inflammatory models. However, the effects of AS on liver injury are poorly understood. The present study was undertaken to investigate whether AS is efficacious against Lipopolysaccharide (LPS) /D-galactosamine (D-GalN)-induced acute liver injury in ...

The Protective Effect of Cordycepin on D-Galactosamine/Lipopolysaccharide-Induced Acute Liver Injury

As the major active ingredient of Cordyceps militaris, cordycepin (3'-deoxyadenosine) has been well documented to alleviate inflammation and oxidative stress both in vitro and in vivo. To explore the potential protective effect of cordycepin in fulminant hepatic failure, mice were pretreated with cordycepin for 3 weeks followed by D-galactosamine (GalN)/lipopolysaccharide (LPS) injection. Then ...

Protective effect of chloral hydrate against lipopolysaccharide/D-galactosamine-induced acute lethal liver injury and zymosan-induced peritonitis in mice.

In recent years, certain anesthetics have been shown to have protective effects against acute inflammation in experimental animals, an observation that may yield new options for adjunctive treatment of acute inflammation. In this study, we investigated the effects of chloral hydrate (CH) on the acute inflammatory response in BALB/c mice using lipopolysaccharide/D-galactosamine (LPS/D-GalN)-indu...

Early growth response-1 contributes to galactosamine/lipopolysaccharide-induced acute liver injury in mice.

Early growth response (Egr)-1 is a transcription factor that regulates genes involved in inflammation, innate and adaptive immunity, coagulation, and wound healing; however, little is known about the role of Egr-1 in acute liver injury. We tested the hypothesis that Egr-1 is involved in acute liver injury induced by galactosamine/lipopolysaccharide (GalN/LPS). GalN/LPS exposure biphasically inc...

Protective Effect of Penta-acetyl Geniposide on Acute Liver Injury Induced By D-galactosamine in Mice

Penta-acetyl geniposide ((Ac)5GP), an herbal derivative prepared from Gardenia Fructus geniposide, decreased the DNA damage and hepatocarcinogenesis induced by aflatoxin B1 (AFB1). The present study was carried out to further evaluate the protective effect of (Ac)5GP on liver injury induced by D-galactosamine (DGalN). D-GalN (400 mg/kg) was given to mice by intraperitoneal injection, while (Ac)...
